Study of the effect of Shen Mai Injection on the inflammation and proliferation of airway smooth muscle cells in asthmatic rats
Objective To observe the effect of the transient receptor potential ankyrin 1(TRPA1)channel on the proliferation and inflammation in asthmatic rat airway smooth muscle cells.Methods Rats were randomly divided into three groups: the control group,the asthma group,and the SMI treatment group,with 15 rats in each group.The asthmatic rat models were induced by inhalation of ovalbumin.The rats of the control group and the asthma group were injected 2mL NS intraperitoneally every day.The rats of SMI treatment group were injected 2mL SMI intraperitoneally every day.The mRNA and protein expression of TRA1 and proliferating cell nuclear antigen(PCNA)were detected by real-time quantitative reverse transcriptase polymerase chain reaction(RT-qPCR)and Western blotting.The expression of interleukin(IL)-4,IL-5 and IL-13 in the serum were detected by enzyme-linked immunosorbent assay(ELISA).Results The expression levels of TRPA1,PCNA mRNA and protein,IL-4,IL-5 and IL-13 in asthmatic rats were significantly higher than those in the control group(P <0.05).After SMI treatment,the expression of TRPA1,PCNA,IL-4,IL-5 and IL-13 were significantly lower than those in asthmatic group(P<0.05).Conclusion TRPA1,IL-4,IL-5 and IL-13 were highly expressed in tracheal smooth muscle cells of asthmatic rats.The inhibition of TRPA1,IL-4,IL-5 and IL-13 might be one of the mechanisms to intervent the chronic airway inflammation and proliferation of asthma by SMI.
