Changes of new epigenetic modifications in the lungs of cigarette smoke extract-induced murine emphysema model
OBJECTIVE: To investigate whether there are a variety of changes in epigenetic modifications in lung tissue of cigarette smoke extract-induced murine emphysema model in order to provide a new target for the treatment of chronic obstructive pulmonary disease(COPD).METHODS: 12 BALB/c mice were randomly divided into 2 groups: CONTROL group,CSE group,with 6 mice in each group.CONTROL group was injected intraperitoneally with 0.3mL PBS at days 0,11,and 22.The other group was injected intraperitoneally with 0.3mL CSE at days 0,11,and 22.The lung function and morphometry were measured.The acetylation,ubiquitination,malonylation,succinylation,crotonylation changes were measured by Western blotting.RESULTS:1.Compared with the CONTROL group,the airway resistance(RAW)of the CSE group was significantly increased and the lung compliance(Cdyn)was significantly decreased.2.Compared with the CONTROL group,the MLI and DI of the lung tissue of CSE group were significantly increased.3.Compared with CONTROL group,acetylation,ubiquitination,succinylation and crotonylation of histone in CSE group were significantly higher than those in control group,and there was no significant difference in malonylation,CONCLUSIONS: There are a variety of new epigenetic changes in the lungs of lung emphysema induced by CSE,but the specific role and mechanism need to be further verified and explored.
