Molecular Cytogenetics,Genome Size of Male and Female Trichosanthes kirilowii Maxim.-a Traditional Chinese Herbal Medicine
An intensive karyotypeanalysis of Trichosanthes kirilowii Maxim.,a Chinese herb in the family Cucurbitaceae,was carried out using fluorescence in situ hybridization.The results showed that T.kirilowii isoctoploid and has 88 chromosomes in somatic cells.The C-value of the female individuals is 5.12 pg/2C,while that of the male individuals is 4.83 pg/2C,with a difference of 0.1G between the males and females.The results revealedthenumberof5SrDNAiseight,and the 45SrDNAsignal is 10.Furthermore,non-integral ploidy was not observed.Genomic in situ hybridizationanalysisfound differences in the hybridization positions.We statistically analyzed the length and arm ratio of the chromosomes and obtained a preliminary formula for the female and male karyotype.The data implied that the karyotype of the T.kirilowii isoctoploid,the genome of the female plant is larger than that of the male plant,and there is no sex chromosome,which makes T.kirilowii a promising model for studying sex chromosome evolution.The karyotype of plants in Trichosanthes were analyzed,and found that the plants were multiplied by 11 as the base number,which laid the foundation for the prediction of the genome size of other plants in the genus;then we analyzed the relationship between the karyotype and geographical distribution,and showed that the low ploidy plants were mainly distributed in the tropical and subtropical regions,while the high ploidy plants were mainly distributed in the temperate zone and the cold zone.Finally,according to the analysis of karyotype of Cucurbitaceae,We carried out a possible explanation for pollination phenomena of Trichosanthes.
