Protective effect of polysaccharides isolated from the seeds of Cuscuta chinensis Lam.on 5-fluorouracil-induced intestinal mucositis in mice
Background:Intestinal mucositis is a serious toxic side effect of 5-fluorouracil(5-FU)treatment.It is reported that appropriate plant-based medicines may provide better treatment options for Intestinal mucositis disease.As a traditional medical agent,polysaccharide from Cuscuta chinensis Lam(PCCL)has been used in numerous human diseases for thousands of years.However,the effect of treatment for intestinal mucositis by using PCCL has not been fully understood.Our research aims to explore the potential effects by using mice models.Methods:MaleC57BL/6 mice were assigned randomly to three groups:control(PBS),5-FU(5-FU),5-FU+PCCL(5-FU+PCCL 20mg/kg).PBS in control group was given the same dose as PCCL in 5-FU+PCCL group.For the 5-FU group,5-FU was injected i.p and once daily at a dose of 50 mg/kg for 5 days(day 2 to 7).5-FU+PCCL groups had doses of PCCL administered i.g 2 days before 5-FU injection began and twice daily for 7 days(day 0 to 7).Diarrhoea score and body weight were assessed.At day 7,the mice were sacrificed and series of steps followed:(1)Histopathology for observing small intestinal villi height and crypt length,(2)Terminal deoxynucleotidyl transferase-mediated deoxyuridine triphosphate nick-end labelling(TUNEL)assay.(3)Measurement of tissue inflammatory factor(tumor necrosis factor a,interleukin 6 and interleukin ip)levels.(4)Immunohistochemistry for Cleaved Capase-3,BAX and BCL-2 in the jejunum.Results:PCCL administration was found to attenuate the severity of mucosal injury significantly(as reflected by body weight changes,degree of diarrhea,height of villi,and damage to crypts).A decrease in the levels of tissue inflammatory factors,the apoptosis rate,and the expression of caspase-3 and Bax were found,whereas Bcl-2 expression increased.Conclusion:PCCL administration may have significant protective effects against 5-FU-induced intestinal mucositis by inhibiting cytokine-mediated apoptosis pathway.
