Chemical Fingerprint and Quantitative Analysis for the Quality Evaluation of Bletilla striata by UPLC-DAD-TOF-MS Coupled with Chemometrics Analysis

  Bletilla striata.root from the genus of Bletilla Rchb.f.is an important traditional Chinese medicine(TCM).However,three species including Bletilla striata(Thunb)Reichb.f.,Bletilla formosana(Hayata)Schltr.,Bletilla formosana(Hayata)Schltr.,which distributed in mainland china,are simultaneous existence in the herb market,then they are often used confusingly and influence clinical therapeutic effect.A rapid and effective method by means of ultra-performance liquid chromatography(UPLC)fingerprint method couped with diode array detector(DAD)combined with chemometrics analysis(including the similarity analysis,hierarchical cluster analysis and partial least-squares discrimination analysis)has been established for quality assessment of Bletilla striata.root.The validated method was successfully applied for discrimination and simultaneous determination of the three chemical constituents in 27 bathces of samples collected from different regions in china.From the chemical fingerprint,19 common peaks were obtained among the 27 samples,meanwhile the chemometrics analysis based on the area of 19 common peaks was successfully distinguish 27 batches of Bletilla striata.root samples from these three species.Furthermore,7 characteristic common peaks were selected as potain chemical markers,which were influenced the quality of these three species.UPLC combined with DAD and time-of-flight(TOF)mass spectrometry(MS)detection and chemometrics analysis was used to tentative identification of 7 characteristic common peaks.The contents of militarine and kaempfer varied remarkably with ranges of 7.86-33.01 and 0.27-2.81mg/mL,respectively.This study demonstrated that the chemical fingerprint combined with chemometrics analysis can efficient evaluation the quality of Bletilla striata.root.
