马红丸通过抑制肝癌细胞的侵袭迁移调节DCD通路的表达

  Background and Aim:Dermcidin(DCD)could interact with NCK1 to activate the Rac1 and cell division control protein 42(Cdc42)to promote the invasion and metastasis of cancer,including human hepatoma.This study aimed at investigating the effect of Mahong,aqueous extracts of eight Traditional Chinese Medicines on DCD-related NCK1,Rac1 and Cdc42 expression,adhesion,migration and invasion of human hepatoma SK-Hep-1 cells in vitro.Methods:The role of Mahong in SK-Hep-1 cells adhesion,migration and invasion capacities was evaluated by using cells adhesion assay and transwell assay in vitro.The expression and distribution of DCD and NCK1 in DCD-silencing and DCD over-expressing SK-Hep-1 cells was observed by confocal immunofluorescent microscopy.Western blot analysis was performed to assess different concentrations Mahong interacting with the DCD/NCK1/Rac1 and Cdc42 signaling pathway.Results:We found that treatment with different doses of Mahong did not alter the viability of SK-Hep-1 cells in vitro.Adriamycin dramatically increased the levels of DCD,but decreased the levels of NCK1 expression in DCD over-expressing SK-Hep-1 cells.In contrast,Mahong at 20%almost completely eliminated the DCD expression and clearly decreased NCK1 expression in DCD-over-expressing SK-Hep-1 cells.Mahong,like Adriamycin,significantly reduced the relative levels of Rac1 and Cdc42 expression in DCD-over-expressing cells and inhibited the adhesion,migration and invasion of SK-Hep-1 cells in vitro.Conclusions:Our data indicated that Mahong potently inhibited the adhesion,migration and invasion of human hepatoma cells by down-regulating the DCD/NCK1/Rac1 and Cdc42 signaling.
