Analysis of Longan POD Isoenzyme Activity
The methods of POD isoenzyme extraction and electrophoresis were optimized.The result showed that 3 to 6 month young leaf was suitable for POD isoenzyme.The extraction buffer contains 0.05 mol/L phosphate buffer (Ph 7.5) and 0.1% TritonX-100. POD isoenzyme was separated by polyacrylamide gel electrophoresis. The zymogram of POD isoenzyme increased from 8 to 17 using improved POD extraction and electrophoresis methods.The zymogram was divided into 4 regions. Region Ⅰ and Ⅲ were active regions. The zymograms in these regions were active and conserved.Region Ⅱ and Ⅳ were weak regions,and the zymograms were weak. It was concluded that the second and third bands were distinct bands.
