Dynamic expression of rat heat shock protein gp96 and its gene during development of hepatocellular carcinoma

Hepatocellular carcinoma (HCC) is characterized by multicause, obvious multistage and multifocus process of tumor progression. Development of HCC is related intimately with overexpression and signal transduction of many cellular factors. The aim of this study was to investigate the dynamic expression and alteration of heat shock protein (HSP) gp96 along with its gene during HCC development. Rat hepatoma models were established by using male S-D rats induced with 2-Fluorenylacetamide. Total RNAs were transcripted into cDNA by reverse transcription and the gene fragment of gp96 was amplified by nested RT-PCR. The gp96 expressions in rat liver tissues were semi-quantitatively analyzed by immunohistochemistry. An increasing tendency of liver gp96 protein was found from normal liver to precancerous to cancerous tissues during rat hepatoma development (P<0.01), which was in accordance with gp96 mRNA alteration (P<0.05). HSP gp96 is involved in HCC development and its overexpression could be useful marker for early diagnosis of HCC.
