Effect of ezogenous cholesterol on cholesterol accumulation and mRNA ezpression of SREBP-2 and HMGR in goose primary hepatocytes
In order to understand the effect of exogenous cholesterol on cholesterol accumulation and mRNA expression of SREBP-2 and HMGR in goose, primary hepatocytes from Landes Goose (Anser anser) and Tianfu Meat Goose (Anser cygnoides) were isolated and treated with the exogenous cholesterol.Cholesterol accumulation, and gene expression levels of SREBP-2 and HMGR were measured.The results indicated exogenous cholesterol cultured with hepatocytes for 72h had no evident effect on TC content in hepatocytes.We found TC content increased before 24h, decreased after 48h.Exogenous cholesterol could decrease the mRNA level of SREBP-2 and HMGR.The correlation analysis indicated the mRNA expression of SREBP-2 was positively correlated to that of HMGR.In conclusion, exogenous cholesterol could regulate the cholesterol content by decreasing the gene expression of SREBP-2 and HMGR in goose primary hepatocytes.
