Resveratrol induces human hepatoma cancer cells HepG-2 apoptosis via mitochondria pathways
To study mechanism of resveratrol induce tumor cell apoptosis through mitochondria pathways. Measurements using mononuclear cell direct cytotoxicity assay shows that its cytotoxic effect on HepG-2 is strong with IC50 being 34.86 μmol·L-1.Measurement of action of apoptosis using morphology with Hoechst 33258 stain show that karyoplasmic ratio of tumor cells diminish, cell nucleus pyconsis, the staining extent become deeper, chromatin prosperous aggregation, fragmentation. The cells breaks up into several apoptotic bodies of different sizes. As RES concentration is increased, these morphological changes under microscope become more and more clear, indicating that the proportion of cells undergoing apoptosis is gradually increasing. Measurement tumor cell cycle dealed with RES using flow cytometry shows that RES leads to a decrease in the proportion of cells in G0/G1 phase, an increase in the proportion of cells in the S phase and decrease in the proportion of cells in G2/M phase.This shows that RES has influence in thetumor cell cycle and can block tumor cells from S phase to G2/M phase.At sametime, The rate of apoptosis is 26.130%,31.599%,65.655%.Measurement mitochondrial membrane electric potential(Δψm) treated with RES using the flow cytometry with Rodanmin 123 stain shows that RES can obviously cut down the Δψm of the tumor cells with the increasement of the dose. This effect shows dose-dependent. This effect shows dose-dependent too.In a word, RES can induce tumor cell apoptosis via mitochondria pathways.
