Microbial Community Structure of Biological Contact Ozidation Process Used in Landscape River

Urban landscape water pollution is widely and serious at present. Biological contact oxidation process was applied to purify the landscape river in this paper. In order to ulteriorly analysis the growth of biomembrane and mechan-ism of water purification process, 16S rDNA and amoA gene based amplification and denaturing gradient gel electrophoresis (PCR-DGGE) were used to study the microbial community structure of biomembrane in biological contact oxidation packing. Biomembrane samples were collected from different date during the biomembrane forming and mature period. Superior bacteria and amount of microbe were both lesser in the earlier forming stage, and genetic similarity between biomembrane forming period and mature period is 45%. Community of mature biomembrane samples was a little different from each other for the diverse water temperature, similarity of the samples of T6 and T7 was the highest, up to 91%. The sequences of amoA gene DGGE fragment were determined, their close relatives in GenBank were uncultured beta proteobacterium from polluted environment with ammonia-oxidizing ability.
