HETEROZYGOTE DETECTION BY COLOR-CODED MINISEQUENCING
Dual-color method was applied in solid phase minisequencing to genotyping SNP, including heterozygote with 2 alleles. But this method could not genotype heterozygote with over 2 alleles. In this study, we introduced color-coded method to solid phase minisequencing to genotype heterozygote with over 2 alleles. In our experiment, 4 templates with 4 different alleles were synthesized. Cy3 - ddUTP, Cy3 - ddCTP, Cy5 - ddUTP, and Cy5 - ddGTP were mixed as the color-coded ddNTPs in our minisequencing experiment. The on-chip extension was performed subsequently with the color-coded ddNTPs. As what we anticipated, the signals of the six templates mixtures in Cy3 and Cy5 channel exhibit evident differences between each other and the results demonstrated the feasibility of 2 - color solid phase minisequencing in heterozygote genotyping.
