Preparation of CNTs/HAp/Chitosan hydrogel and Its Cellular Compatibility
This study is made to prepare CNTS/HAp/CS thermosensitive hydrogel and detect its cytotoxicity according to the national standards in order to provide a new and effective thermosensitive hydrogel for tumor thermotherapy for clinical practice. Firstly, Nano-HAP powder prepared through a chemical precipitation process is mixed with CNTS in a mass ratio of 1:1 to obtain Hap-coated CNTs by 6h ball milling under wet conditions at the rate of 300r/min,which is then surface modified by chitosan. The chitosan- Sodium glycerophosphas is added with citric acid of certain content to get low-pH thermosensitive hydrogel which is uniformly mixed with HA/CNTs powder by some specific technique to acquire CNTS/HAp/CS thermosensitive hydrogel. Secondly, four L929 cell-culture plates are respectively put into 100μL extract solutions of different concentrations(at 100%、75%、50% and 25%) for 72hours. The density and morphology of cells are observed, RCGs are detected with MTT assay and cytotoxicities of materials are measured. And finally, 3mm-diameter disks processed by thin film materials of low temperature and drying are co-cultured with L929 for 72, 120 hours to prepare electron microscopic samples. The relationship between cell morphology and materials are studied. The cellular toxicity of prepared materials range from 0 to 1 degree with little effect on proliferation and morphology of cultured cells. The results show this material has good cellular compatibility as biomedical material.
