MEASUREMENT OF WATER TRANSPORT CHARATERISTICS OF BONE MARROW STROMAL STEM CELLS

The methods for the cryopreservation of bone marrow stromal stem cells have largely been developed empirically, but may require refined freezing processes to achieve optimal recovery after freezing. To predict optimal freezing processes for cryopreservation of pig bone marrow stromal stem cells, knowledge of fundamental cryobiological characteristics including cell osmotic characteristics is required. In this study, the osmotic characteristics of pig bone marrow stromal stem cells were experimentally measured using electronic sizing technique. The cell volume measured is 5248.4μm3,the relative diameter 21.6μm,at isotonic osmolality. The cell volume change was measured after the cells exposed and equilibrated to different anisosmotic conditions. The cell volume was found to be a linear function of the reciprocal of the extracellular osmolality (Boyle van’t Hoff plot) .The non-osmotic volume of the cell is 36% of the cells isotonic volume.
