Crystalline Silica Promotes Rat Fibrocyte Differentiation in Vitro and Fibrocyte Participates in Silicosis in Vivo
Fibrocytes are known to derive from bone marrow and to be involved in the pathogenesis of fibrosis diseases.While crystalline silica induced fibrocyte differentiation is less well studied.We found that peripheral blood monocytes differentiate into fibrocytes in vitro by time dependent manner and crystalline silica (1 O0 μg/ml)exposure may potentiate fibrocytes differentiation.Furthermore, TGF-β1 increased is associated with the differentiation of cultured fibrocytes.In addition,fibrocytes can migrate to lung tissue at day 7 after silica exposure by flow cytometry and immunofluorescence.We also observed fibrocytes in lung tissue after 1 week and 6 weeks crystalline Silica exposure.Taken together, these data shows that peripheral blood monocytes can differentiate into fibrocytes in vitro and Silica (lOOμg/ml)exposure may potentiate fibrocytes differentiation.Fibrocytes are involved in the pathogenesis of silicosis.
