THE Inhibiting Effect of EZH2-targeting shRNA on SW480 Cells and the Impact of 5-FU on this Inhibition
Aim: To investigate the inhibiting effect of EZH2-specific shRNA on human colon cancer cell SW480 and the impact of 5-FU on this inhibition.Materials and Methods: SW480 cells were transfected with EZH2-specific shRNA (SW480-EZH2-shRNA).The expression of EZH2mRNA and protein, and the expression level of EZH2 mRNA by tumor tissues were detected by PT-PCR and Western blot.Results: Compared with the negative control group, the expression of EZH2mRNA and protein of gene silencing (GS) group were both obviously decreased (P<0.01 and P<0.05, respectively).In nude mice, tumor volume and weight of EZH2 mRNA expression in 5-FU-GS group and GS group were all decreased compared with that of normal group.Conclusion: The EZH2-targeting shRNA can inhibit EZH2 mRNA and protein expression in SW480 cells.In vivo, 5-FU can inhibit EZH2 mRNA expression in EZH2-GS SW480 cells.The paper lays foundation for further exploration of EZH2 inhibition on tumor cells.
